Dielaidoylphosphatidylethanolamine (DEPE) is a synthetic phospholipid featuring trans-unsaturated acyl chains, commonly used as a model membrane lipid for studying polymorphic phase behavior. This compound undergoes a characteristic gel-to-liquid crystalline (Lβ to Lα) transition near 37-38°C and a lamellar-to-inverted hexagonal (HII) phase transition at approximately 63°C, making it a standard system for investigating non-bilayer lipid structures. DEPE is widely employed in biophysical research to examine how various additives—such as ether lipids, biosurfactants, or short-chain alcohols—modulate membrane curvature and phase stability, with calorimetric and NMR data showing that concentrations above 5 mol% of certain amphiphiles can completely suppress the HII phase formation.
