Introduction

The Human Cytokine Array C5 is a multiplex, membrane-based antibody array designed for simultaneous screening of 80 human cytokines from a single sample. This high-density cytokine profiling platform combines sandwich ELISA specificity with the multiplex capability of protein arrays, enabling rapid comparison of cytokine expression patterns across experimental conditions.

Unlike traditional single-target ELISA assays or complex bead-based multiplex platforms, the Cytokine Array C5 provides a simple western blot-like workflow that can be completed in one day with minimal optimization. The array is compatible with serum, plasma, cell culture supernatants, tissue lysates, and cell lysates, making it ideal for biomarker discovery, inflammation research, and pathway analysis.

Typical Data

RayBiotech’s Human Cytokine Array C5 (AAH-CYT-5) was used to evaluate the cytokine response of bronchoalveolar lavage macrophages following stimulation with extracellular nucleosomes. Increased secretion of TNFα, IL-6, IL-10, CCL7, CCL8, CXCL9, and CXCL10 was observed 4H post stimulation. Figure adapted from Sripada et al., J Clin Invest, 2025.

RayBiotech’s Human Cytokine Antibody Array C5 (Cat# AAH-CYT-5) was used to profile SASP factors in conditioned media from WT and ATG7 KO valvular interstitial cells, with differential cytokine expression visualized as a heatmap. Figure adapted from Tang et al., Autophagy, 2025. .

AAH-CYT-5 was used to assess cytokine secretion in conditioned media from human dermal papilla cells, identifying reduced levels of IL-6, IL-8, MCP-1, and other senescence-associated factors following senolytic treatment. Figure adapted from Pappalardo et al., Aging Cell, 2025.

Cat# AAH-CYT-5 was used to assess cytokine secretion in conditioned media from human dermal fibroblasts, identifying osteoprotegerin (OPG) as a TGF-β1-induced factor associated with pro-fibrotic signaling. Figure adapted from McDowell-Sanchez et al., Biochemical and Biophysical Research Communications, 2025.

Cat# AAH-CYT-5 was used to assess cytokine secretion from macrophages exposed to tumor cell and mesenchymal stem cell conditioned media, identifying increased levels of CXCL1, CXCL5, and CXCL13 associated with stromal cell–driven immune modulation. Figure adapted from Zhang et al., Communications Biology, 2024.

Cat# AAH-CYT-5 was used to assess cytokine secretion in conditioned media from proliferating and senescent fibroblasts, identifying increased levels of BDNF, IL-6, CXCL1, HGF, and CXCL10 associated with the senescence-associated secretory phenotype. Figure adapted from Anerillas et al., Nature Communications, 2022.

Cat# AAH-CYT-5 was used to assess cytokine secretion from E. coli–activated human breast fibroblasts, identifying increased levels of inflammatory and pro-carcinogenic cytokines including IL-1β, IL-6, IL-8, MCP-1, and GRO family chemokines. Figure adapted from Alshehri et al., Cells, 2024.

Cat# AAH-CYT-5 was used to assess cytokine secretion in conditioned media from SKAP1-overexpressing colon cancer cells, identifying CXCL8 as the most strongly upregulated factor associated with neutrophil recruitment and tumor-promoting immune modulation. Figure adapted from Gao et al., Advanced Science, 2024.
