AMPK is a highly conserved heterotrimeric protein kinase found in all eukaryotes. The heterotrimeric complex exists as multiple isoforms and consists of a catalytic alpha subunit, which is stabilised by non-catalytic regulatory beta- and gamma- subunits. AMPK plays a vital role in maintaining cellular energy homeostasis and is activated by rising levels of AMP under conditions of hypoxia, ischemia, exercise, low glucose levels.
AMPK may play an important role in the control of type 2 diabetes and metabolic syndromes. Recent studies suggest that AMPK functions as a suppressor of cell proliferation of both malignant and normal cells.
Applications:
Suitable for use in ELISA, Immunofluorescence, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested.
Recommended Dilutions:
Western Blot: 1:100-1:1,000. Detects a band of ~63kD in rat brain cell lysates.
Immunohistochemistry: Frozen sections
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
