Cell Detachment Procedure
Work aseptically in a laminar flow hood.
1. Remove medium; rinse with calcium- and magnesium-free salt solution; aspirate.
2. Prewarm reagent to 37°C; cover cells.
3. Incubate at 37°C; sensitive cultures: room temperature or 2–8°C.
4. Monitor rounding and cloudiness frequently; minimize exposure to prevent damage.
5. Neutralize with serum-containing medium or trypsin inhibitor; gently centrifuge; discard supernatant.
6. Resuspend in fresh medium; count or culture.
Detachment time varies with cell type, culture age, density, serum concentration, and time since subculture.
