Tryptic soy broth is a growth medium used in the cultivation of bacteria. It contains tryptone, yeast extract, and sodium chloride. Tryptic soy broth is used for the isolation of type strains and blood culture specimens to identify Staphylococcus species. The pH of the medium is adjusted with hydrochloric acid or sodium hydroxide before autoclaving to eliminate any residual bacteria that may have contaminated the ingredients. The final pH should be between 6 and 7.5 for optimal bacterial growth. Tryptic soy broth contains penicillin-binding protein (PBP) which helps to inhibit bacterial growth by binding to it, preventing bacterial cell division from occurring.
