Dihydroxyacetone (DHA), also known as glycerone or 1,3-dihydroxy-2-propanone, is the simplest ketotriose monosaccharide with the chemical formula C3H6O3. It is a small, three-carbon sugar characterized by a ketone functional group on the second carbon and hydroxyl groups on the first and third carbons. Unlike aldehyde-containing sugars, DHA has no chiral centers, making it achiral.

Chemically, dihydroxyacetone commonly exists as a hygroscopic white crystalline powder that easily forms a dimeric structure (2,5-bis(hydroxymethyl)-1,4-dioxane-2,5-diol) in solid form. When dissolved in water, the dimer dissociates to the monomeric form, which is stable in mildly acidic to neutral pH conditions (around pH 4–6), but degrades in more basic environments. It has a melting point around 89–91°C and exhibits a sweet cooling taste with a characteristic odor.

Biological and Industrial Importance

Biologically and industrially, DHA is widely known for its application in sunless tanning products. Its mechanism of action involves a Maillard reaction between DHA and amino acid residues of keratin in the skin’s outer layer, leading to the formation of melanoidins — brown-colored polymers that mimic natural tanning pigments without UV exposure. This reaction requires oxygen and does not occur with injection, emphasizing its surface-level interaction when applied topically.

DHA is primarily derived from natural sources such as sugar beets and sugar cane, or produced through glycerin fermentation. Beyond cosmetics, DHA plays various biochemical roles and serves as an essential metabolic intermediate, particularly in its phosphorylated form — dihydroxyacetone phosphate — within glycolysis and other metabolic pathways.
