ELISA Kit Principle | Aviva Systems Biology 2, 3-Bisphosphoglycerate ELISA Kit (OKEH02577) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-2, 3-Bisphosphoglycerate antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated 2, 3-Bisphosphoglycerate and incubated. The 2, 3-Bisphosphoglycerate found in the sample or standards competes with the biotinylated 2, 3-Bisphosphoglycerate for limited binding sites on the immobilized anti-2, 3-Bisphosphoglycerate antibody. Excess unbound biotinylated 2, 3-Bisphosphoglycerate and sample or standard 2, 3-Bisphosphoglycerate is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated 2, 3-Bisphosphoglycerate captured in the well and inversely proportional to the amount of 2, 3-Bisphosphoglycerate which was contained in the sample or standard. | ||||||||||||||||||||||

Description of Target | 2,3-Bisphosphoglyceric acid (2,3-Bisphosphoglycerate or 2,3-BPG, also known as 2,3-diphosphoglycerate or 2,3-DPG) is a three-carbon isomer of the glycolytic intermediate 1,3-bisphosphoglyceric acid (1,3-BPG). 2,3-BPG is present in human red blood cells (RBC; erythrocyte) at approximately 5 mmol/L. It binds with greater affinity to deoxygenated hemoglobin (e.g. when the red cell is near respiring tissue) than it does to oxygenated hemoglobin (e.g., in the lungs) due to spatial changes: 2,3-BPG (whose size is estimated at about 9 angstroms) fits in the deoxygenated hemoglobin configuration (11 angstroms), but not as well in the oxygenated (5 angstroms). It interacts with deoxygenated hemoglobin beta subunits by decreasing their affinity for oxygen, so it allosterically promotes the release of the remaining oxygen molecules bound to the hemoglobin, thus enhancing the ability of RBCs to release oxygen near tissues that need it most. 2,3-BPG is thus an allosteric effector. |
