4°C Do Not FreezeRecognizes a conserved determinant of the p53 protein. Its epitope maps near the C-terminus end (aa 370-378) of p53. It has been used successfully to immunoprecipitate p53 oncoprotein complexes with simian virus 40 (SV40) large T antigen. Reportedly, this antibody precipitated the most ATPase-active form of T. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.
Applications:
Suitable for use in Immunofluorescence, ELISA, Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 0.5-1ug/million cells
Immunofluorescence: 0.5-1ug/ml
Western Blot: 0.5-1.0ug/ml
Immunoprecipitation: 0.5-1ug/500ug protein lysate
Immunohistochemistry: Frozen only: 0.5-1.0ug/ml for 30 minutes at RT
Optimal dilutions to be determined by the researcher.
Positive Control:
MDA-MB-231 Cells. Breast or Colon carcinoma
Storage and Stability:
May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer. APC is light sensitive.
Note: Applications are based on unconjugated antibody.
