Serum-free cryopreservation media are specialized formulations designed to preserve cells and tissues at ultralow temperatures without the use of animal or human serum. Traditional cryopreservation protocols have relied on fetal bovine serum (FBS) as a key component, typically at concentrations of 20–90% combined with 5–10% dimethyl sulfoxide (DMSO). However, FBS introduces substantial scientific, regulatory, and ethical challenges: its composition varies greatly between lots, it carries risks of contamination with viruses, prions, and mycoplasma, and it contains undefined bioactive factors that can unpredictably affect cell physiology. Serum-free cryopreservation media address these limitations by replacing serum with defined or semi-defined components that provide cryoprotection, osmotic support, and membrane stabilization without the drawbacks associated with animal-derived materials.
Applications
Serum-free cryopreservation media are particularly critical in cell and gene therapy manufacturing, where regulatory requirements demand defined, animal-free production systems. The rapid clinical translation of immune cell therapies necessitates cryopreservation strategies that preserve post-thaw viability and functional integrity. Conventional 10% DMSO-based, serum-containing formulations induce cryoinjury by disrupting membrane integrity, altering cytokine and chemokine signaling, and reducing immune effector function. These toxicities, coupled with safety, regulatory, and scalability limitations, constrain large-scale manufacturing and direct clinical infusion.
Low-DMSO, serum-free, osmoprotectant-supplemented GMP-compliant customized cryopreservation formulations have been tested as viable alternatives. Tailored cell type-specific cryoprotectant strategies have been optimized: NK cells benefit from low-DMSO, trehalose-supplemented formulations that preserve granzyme B/perforin expression and IFN-γ/TNF-α secretion; CAR-T cells maintain TCR signaling and IL-2/IFN-γ production in serum-free, proline-enriched media; and dendritic cells retain IL-12/IL-10 secretion and co-stimulatory molecule expression in low-DMSO, antioxidant-supplemented CryoStor formulations.
