Verified Activity
Immunofluorescence staining of TBXT in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with mouse anti-Human T monoclonal antibody (dilution ratio 1:60) at 4°C overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-mouse IgG secondary antibody (green). Positive staining was localized to Nucleus.

Research BackgroundBrachyury is encoded by the TBXT gene. TBXT plays in the Cnidaria, appears to be in defining the blastopore. Brachyury is an important factor in promoting the epithelial-mesenchymal transition (EMT). Cells that over-express brachyury have down-regulated expression of the adhesion molecule E-cadherin, which allows them to undergo EMT. This process is at least partially mediated by the transcription factors AKT and Snail. Brachyury is overexpressed in a number of tumor types.
