-20°CHuman CD81 is a member of the tetraspan family of leukocyte cell surface molecules which are involved in signal transduction, cell-cell adhesion and cellular activation. Association of CD81 with integrins appears to be functionally important in the regulation of cell adhesion and activation. Recent work with CD81-null mice indicates CD81 is important for development of humoral immune response.
Applications:
Suitable for use in Flow Cytometry, Immunocytochemistry and Western Blot. Other applications not tested.
Recommended Dilutions:
Flow Cytometry: 10ug/ml; 80ul labels 5x10e5 cultured human Raji cells.
Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol:
1. 5x10e5 cultured human Raji cells per tube were incubated for 45 minutes on ice with 80ul of C2433-10B at 10ug/ml.
2. Washed cells twice and incubated with S7973-88P: Streptavidin (PE).
3. Washed cells three times, fixed and analyzeed by FACS.
4. Cells stained positive with a mean shift of 2.3 log10 fluorescent units when compared to IgG1 Murine Negative Control (Biotin) at a similar concentration.
5. Binding was blocked when cells were pre-incubated with 20ul of C2433-10A at a concentration of 0.5mg/ml.
Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
