| Characterization | To ensure acceptable performance, each batch of product is tested in a sandwich ELISA to conform tocharacteristics of a standard reference reagent. |
| Warning | Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact withskin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, washwith copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azideyields toxic hydrazoic acid under acidic conditions. Dilute azide- containing compounds in running waterbefore discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing. |
| Application Info | ELISA Detection: MP4-25D2 monoclonal antibody is useful as a detection antibody in a sandwich ELISA for quantifying human IL-4 protein levels. 1,3 Biotinylated MP4-25D2 antibody should be paired with a suitable purified antibody as the capture antibody, with purified recombinant human IL-4 as the standard. Immunofluorescence/Flow Cytometry: MP4-25D2 antibodies are useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-4-positive cells within mixed cell populations. Recombinant human IL-4, unlabeled MP4-25D2 (OASB01219), or Biotinylated Rat IgG1 (OASB00091) should be used as a specificity control. Western Blotting: The purified MP4-25D2 antibody is also useful for Western blotting applications. A concentration of 1-5ug/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig, OASB01897) has been found to enable detection of <= 100 ng/lane of recombinant human IL-4 under reducing conditions. Neutralization: The LE/AF form of MP4-25D2 can be used for neutralizing human IL-4 bioactivity. A suitable LE/AF Rat IgG1 (OASB00095) should be used as an isotype control. Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications. |

| Protein Name | interleukin-4 |
| Description of Target | The protein encoded by this gene is a pleiotropic cytokine produced by activated T cells. This cytokine is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. This gene, IL13 and IL5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. |
