CD61 is present on the megakaryocyte/platelet lineage, granulocytes, cells from the monocyte/macrophage lineage and endothelial cells (Moreno et al., 2002), but not by other hematopoietic cells. CD61 is also broadly expressed on tissues, such as epithelial cells from tubules in the kidney (Piriou-Guzylack et al., 2008), spleen, intestinal mucosa and Leydig cells in testis (Moreno et al., 2002).
The epitope recognized by this antibody is not sensitive to EDTA.
Applications:
Suitable for use in Flow Cytometry and Western Blot. Other applications have not been tested.
Recommended Dilutions:
Flow Cytometry: Neat-1:10; 10ul labels 10e6 cells in 100ul.
Western Blot: Detects a band at ~85kD in porcine platelet lysates
Optimal dilutions to be determined by the researcher.
Hybridoma:
Sp2/0 - Ag14 myeloma cells with spleen cells from Balb/c mice.
Recommended Negative Control:
I1904-78R3: IgG1 Murine Negative Control (FITC)
Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
