Our rabbit ChAT polyclonal antibody from PhosphoSolutions is produced in-house. It detects bat, chicken, feline, guinea pig, human, mouse, non-human primate, and rat ChAT and is neat serum. Works in ELISA, WB, IHC, IP.
Immunohistochemistry detection of ChAT in normal cerebral cortex tissue using Anti-Choline Acetyltransferase (ChAT) Antibody (Cat. 316-ChAT). Tissue was mounted and pariffin fixed. Heat induced antigen retrieval was performed using citrate buffer, heated, and then blocked. After washing, the primary antibody (Cat. 316-ChAT) was incubated with the sample at 1:100. Immunoreactivity is seeing as a staining pattern on the cell body, dendrites and axons of neurons.

ChAT
Choline acetyltransferase is a neuronal enzyme which catalyzes the reaction between Acetyl CoA and choline resulting in the formation of acetylcholine. It is therefore found primarily in cholinergic neurons making it a valuable marker for diseases associated with decreased cholinergic function such as Schizophrenia, Alzheimer's disease (AD) and Down syndrome (Holt et al. 1999). Decreased choline acetyltransferase activity in particular has been shown in Schizophrenic subjects (Karson et al 1993). It has furthermore been demonstrated that in patients with AD, there are significantly lower levels of cortical ChAT that correlate with severity of the disease as measured by loss of neuropsychological function (Baskin et al. 1999).

Recommended that the undiluted antibody be aliquoted into smaller working volumes (10-30 uL/vial depending on usage) upon arrival and stored long term at -20° C or -80° C, while keeping a working aliquot stored at 4° C for short term. Avoid freeze/thaw cycles. Stable for at least 1 year.

Antibody against ChAT for use in IHC(P), IP and WB. IP: 1:200 - immunoprecipitates 68 kDa active ChAT from brain and placenta; also precipitates a 27 kDa protein co-expressed with ChAT (lacking ChAT activity) in some cholinergic CNS neurons. IHC(P): 1:100 with no antigen retrieval using IHC-Select Detection with HRP-DAB; immunoreactivity seen on cell body, dendrites and axons (brown). Control tissue: cerebral cortex. Routinely evaluated by IHC-P on normal cerebral cortex. Reactivity with other species not confirmed by IHC. Concentration lot-specific - refer to COA.
