Centrifugal filter units are essential ultrafiltration devices widely used in biochemistry and molecular biology. They enable rapid sample concentration, desalting, and purification by applying pressure-driven membrane separation to retain macromolecules while removing small solutes.

Design and Functionality

These devices typically use a dead-end filtration configuration with a semi-permeable membrane such as regenerated cellulose, polyethersulfone (PES), or similar materials. The membrane is mounted inside a centrifuge-compatible tube and operated at centrifugal forces generally between 2,000 and 5,000 × g, processing sample volumes ranging from 0.5 to 20 mL.

Centrifugal filter units are available with different nominal molecular weight cut-offs (MWCO), commonly between 3 and 100 kDa. These thresholds allow selective retention of target biomolecules such as proteins, antibodies, or nucleic acids while permitting salts, metabolites, and detergents to pass through. Low-binding membrane surfaces are often used to maintain recovery rates above 90–95%, and vertical or horizontal filtration formats help minimize hold-up volumes, sometimes below 50 µL.

Biochemical Applications

In proteomics workflows, centrifugal ultrafiltration devices are used to concentrate dilute proteins, antibodies, or enzymes by 20–100 fold within approximately 15–30 minutes. They also support buffer exchange prior to crystallization experiments or mass spectrometry analysis after enzymatic digestion.

In molecular biology, these units facilitate plasmid DNA cleanup, next-generation sequencing library desalting, and primer removal following PCR amplification. Compared with ethanol precipitation methods, centrifugal filtration preserves higher sample yields without the need for chaotropic reagents. Additionally, they are useful for clarifying cell lysates, fractionating viral particles or extracellular vesicles such as exosomes, and removing unincorporated labels in radiolabeled assays.
